INTERESTS

I am interested in merging sustainable development and materials’ science and in particular in studying biological amphiphiles (known as biosurfactants), from their self-assembly properties in water to the developement of « green » soft materials. Some of the topics that I develop in my research and related characterization tools are given below. Niki BACCILE is a physicochemist working in the field of Soft Matter and Materials’ Science. He obtained a Master 2 (« Laurea ») from the University of Padova (Italy) in 2002 and a DEA (Diplôme d’Études Approfondis) from the Université Pierre et Marie Curie in 2003.

He obtained his PhD in Materials’ Chemistry in 2006 at the Laboratoire de Chimie de la Matière Condensée de Paris, Université Pierre et Marie Curie, Paris (France), under the supervision of Dr. F. Babonneau. The title of his dissertation was Contribution à l’étude des interactions tensioactif/silice au sein de poudres mésostructurées de silice.

As a Post-Doc, he worked at the Institut Charles Gerhardt in Montpellier (France) in 2007 under the supervision of Dr. C. Gérardin and working on the development of mesoporous silica templated by reversible polyion complex micelles. He then worked as a Post-Doc at the Max Planck Institute of Colloids and Interfaces, Potsdam-Golm (Germany), in 2008 under the supervision of Prof. M. Antonietti and working on the synthesis and structural characterization of hydrothermal carbons from renewable resources.

Since 2008, he is full-time CNRS (Centre National de la Recherche Scientifique) researcher (Chargé de Recherche) at the Laboratoire de Chimie de la Matière Condensée de Paris, Sorbonne Université, Paris (France). He currently explores the self-assembly properties of microbial biosurfactants (sophorolipids, glucolipids, cellobioselipids).

He is trained in chemical synthesis and physicochemical characterization of materials and colloids in solutions: XRD, SEM, TEM, cryo-TEM, BET, SAXS, SANS, SLS, DLS, Rheology, solution NMR, solid-state NMR.

pH-dependent Self-Assembly of Functional Glucolipid microbial Amphiphiles

Amphiphilic (water-loving/water-hating) molecules in water self-assemble into a broad variety of morphologies, of which the most important ones are micelles, vesicles and lamellae. Surfactant self-assembly generates a broad interest across many fields of research for its implication in chemistry, physics, materials’ science but also life science and medicine. Amphiphiles are everywhere, from everyday consumers’ products (emulsions, shampoos, detergents, cosmetics) to high-end products like drug carriers (liposomes) to living organisms (cell membrane walls). However, the most fascinating aspect, despite heavy worldwide research since the 50’s, is the unpredictable character of the self-assembly properties of a random amphiphile. Much is understood and well-explained of molecules with a classical hydrophilic (charged or neutral) headgroup and hydrophobic tail but prediction of self-assembly is still not generalized.

Biosurfactants are naturally occurring amphiphiles produced by microorganisms through fermentation in the presence of glucose and vegetable oils. Typical examples are sophorolipids, rhamnolipids or cellobioselipids. These compounds are of broad interest. The general consensus behind their development is that their fully biobased origin, the lack of chemical synthetic step involved in their production, their poor cytotoxicity, if no cytotoxicity at all and their biodegradability. Nonetheless, biosurfactants may have complex chemical structures and their self-assembly properties in water, necessary to know to develop any type of application, are poorly known and hard to predict. This is also verified for the « simplest » structures known, that is glycolipid biosurfactants.

Since 2008, in collaboration with the InBio.be (Centre for INdustrial BIOtechnology and BIOcatalysis) at Ghent University (Belgium), I study the self-assembly properties of a number of microbial biosurfactants produced by both wild type and engineered strains. I employ stimuli-resolved in situ Small Angle X-ray Scattering (SAXS) performed at Synchrotron facilities (ESRF France, Soleil France, Diamond UK, BESSYII Germany) to follow, in real-time with a millisecond-to-second resolution the evolution, the supramolecular morphologies of microbial biobased amphiphiles. I systematically cross these data with experiments performed ex situ using cryogenic Transmission Electron Microscopy, collected at our Lab in colaboration with Patrick Le Griel et Dr. Gervaise Mosser. Additional data on these systems are collected, whenever possible, using neutron scattering, recorded at Institut Laue Langevin in Grenoble (France).

In this way, I studied the self-assembly properties in solution of acidic sophorolipids (C18:1, C18:0, branched, acetylated), glucolipids (C18:1, C18:0) and cellobioselipids and found a number of different self-assembled structures (micelles, vesicles, nanoribbons, lamellae) controlled mainly by pH and of which I could identify the structure (e.g., « Coffee-bean like » micelles) as well as the phase transitions. Recently, a new glycolipid amphiphile displayed a complex self-assembly behavior with at least nine different phases, including a surfactant-like, lipid-like and fibrillator-like behaviour.

Microbial glycolipids have a strong pH-dependent self-assembly process. Most of them form micelles at basic pH, but at acidic pH the variety of structures strongly depend on the number of glucose units and unsaturation. Polymers and biopolymers are present in a countless number of consumer’s products, from housecare to paints, from pharmaceuticals to oil industry, from food science to more exotic nanomaterials. The study of intreractions between polymers and surfactants is an old field of research, especially when polymers and surfactants are oppositively charged. In this case, polymers act and polyions and their interactions with surfactants can be strong and they can modify the surfactant’s phase behaviour in an unpredictable way.

The study of the interactions between microbial biosurfactants and(bio)polymers is a new exciting field of research, because one can expect to prepare fully biobased and biodegradable, although functional and interesting, formulations. This is the Graal of a safe-by-design green chemistry approach. Nonetheless, microbial biosurfactants being more complex molecules than classical surfactants, their interaction with (bio)polymers must be explored from scratch, which is the goal of this research direction.

I have so far explored the interactions between sophorolipids and glucolipids with a set of synthetic (polyethyleneimine, polyallylamine) and biobased (polylysine, chitosan) polymers. The interactions strongly depend on the respective charge, on the nature of the surfactant’s phase but also on the type of phase transition. One can obtain complex coacervates and multilamellar wall vesicles but, in some specific cases, the behaviour is entirely orthogonal, that is surfactant self-assembly occurs independently from, and it is not influenced by, the presence of the polymer. Similarly, multilamellar wall vesicles are only obtained by a phase transition from complex coacervates but not directly.
Similar results were obtained on interpenetrated biopolymerbioamphiphile gels.

Control of the self-assembly properties of amphiphiles paves the way to the exciting field of soft materials, like gels, emulsions, foams. In particular, I explore the hydrogel-forming properties of microbial amphiphiles through their hability to self-assemble. The nature of the self-assembled structure strongly impact the type of gel: saturated double-sugar (sophorose) microbial glycolipids form nanoribbons, which entangle in water to form fibrillar hydrogels. In the meanwhile, a similar compound, containing only one single sugar, stabilizes single lipid sheets, which are able to form another class of much rarer hydrogels: lamellar gels, of which the elasticity is lonely controlled by the density and type of defects in the lamellar stacking of the sheets. Structural studies combining rheology, SAXS, rheo-SAXS at ESRF synchrotron but also neutron scattering and optical polarized microscopy contributed to determine, at best, the structure of these complex materials, which could also be transformed in soft « tough » foams through a freeze-casting process.

In recent work, it was shown that some bioamphiphiles show metallogelation, either through wormilike structures or an unusual fibrillar « nano-fishnet » structure, conferring impressive stability to the gels. Cation-resolved in situ SAXS experiments helped understanding the metallogelation mechanism of a system with promising depollution properties.

The intrinsic nature of amphiphilic molecules is to settle at interfaces, liquid-air, solid-liquid, solid-air, etc… The study of biological amphiphiles at interfaces is long-date topic for the interest of these molecules to stabilize foams or emulsions. However, most approaches in this field have been empyrical, while the molecular, local, approach, developed for classical amphiphiles since long time, is only much more recent.

We study molecular interfaces constituted by biological amphiphiles in different domains, from the stabilization of mineral nanoparticles to the development of Self-Assembled Monolayers (SAMs) to the study of intermolecular forces in lamellar lipid systems. Microbial glycolipids are able to anchor to soft and solid interfaces often through their COOH end-group, may it be left as such or modified by another chemical group (e.g., catechols, thiols). Exposing the sugar headgroup towards the outside generates a number of advantages with respect to the bare interface, like improvement of colloidal stability in water, antiadhesion and antibacterial properties.
